Quick test method mycotoxin Aflatoxin

1.The purpose of use with fast aflatoxin test kits – Mycotoxin

RIDA ® QUICK Aflatoxin RQS is quantitative methods (reviews with the RIDA ® QUICK Aflatoxin RQS) try immune ngiệm on marker to detect aflatoxin in maize.

*Aflatoxins are secondary metabolites of fungi species in Aspergillus flavus, parasiticus and nomius. The fungus occurs in the tropical humidity and due to the contamination of plant food in the water are cultivated. Aflatoxin belongs to the strongest occurring substance.

2.Principle of the test

The test facility is based on immunity on stick, on the stick that contains the Antigen-antibody reaction. Specific antibody against toxin Aflatoxin realized aflatoxin in the molecules in the sample. The results are evaluated and read quickly by RIDA ® QUICK Aflatoxin RQS. The control line is not affected by Aflatoxin in samples and are in all cases to prove the test strip is valid.

It also can use, Freeze-dried fossils-phase disc available Compact Dry to analyze toxic mold speed

3.Reagents (According to kit)

Each kit contains sufficient reagents for 20 the post test. Each test contains:20 stick and a bottle of solvent (4,5ml).

4.Required tools and chemicals (not included in the accompanying)

4.1.The tool

  • Sample crushing machine
  • Weight
  • Shaking machine( options)
  • Filter paper
  • Making tube
  • Pipet 50-100 µl
  • Quick Analyzer toxins mold

4.2. Chemicals

  • Methanol (70%) necessary to adjust the precision.
  • Ethanol alcohol (50%) necessary to adjust the precision.

5.Warning and precautions for user

The post test should only be performed by trained staff. The manual must be taken seriously. Vietnam cable television

The stick is very sensitive to moisture. Stick moisture can affect badly to the post test results. So the test electrodes should be stored in the environment preservation, and use the stick immediately when opening the packaging.

6.Preservation and storage

Archives and preservation kit at temperatures from (2-8theC / 35-46the F).Not preserved the sound test in freezing temperatures.

For most of the time use kit will not ensure the quality of the test results.

No exchange between the solvent reagents among different kit batches .

7.Sample preparation – Quick test method mycotoxin Aflatoxin

Use a solvent for the preparation of sample to room temperature (20-25theC/68-77theC) before using the.

The samples must be stored in a cool place, avoid direct light. The form must be troonj both before taking steps to check.

7.1 corn

  • USDA/GIPSA with extraction of ethanol alcohol
  • For 50 g sample of corn were milled into the appropriate container
  • Transfer 50 g form in the jar mix.
  • Add 100 ml of ethanol alcohol(50%).
  • Mix the samples in 2 minutes at high speed.
  • Then listen to the solution in time 1 minutes.
  • 1.0 ml mixing samples into the tubes get centrifuge.
  • Centrifuge model in a minute .
  • Using the pipet retrieved 200 µ l of solvent (included with kit) and 100 µ l sample after the centrifuge on the new centrifuge tubes (Note each sample should use 1 new pipet )
  • Shake well pipe about 5 times and then use the 100 µ l for posting test.

Sample preparation by methanol or ethanol

  • Take about 10 g template was mashed into the appropriate jar and add 20 ml methanol or Ethanol 20 ml (50%)
  • Close the tube and shake the sample tube of about 5 minutes (by hand or with a shake)
  • To listen the solution in 3 minutes, filter or centrifuge ( This is required to make methanol evaporates)
  • Use 100 µ l of solvent ,Add 50 µ l grade separation on the aqueous portion of the sample canister. stir the mixture and take out 100 µ l for posting test.
  • Notes: If necessary the sample weights can change. And the ratio of the volume of methanol or ethanol according to the ratio

For example:

5g in 10 ml methanol(70%) or 10 ml ethanol (50%).

8.Perform test mycotoxin Aflatoxin

  • Apply with the amount of sample is 100 µ l on stick.
  • Read the result after 5 minutes with the RIDA ® QUICK Aflatoxin RQS
